Nanoparticles in pregnancy: the next frontier in reproductive therapeutics

Pre-clinical research in the development of nanoparticle therapeutic delivery is being undertaken in many fields of reproductive medicine. Non-hormonal-targeted nanoparticle therapy for fibroids and endometriosis may provide fertility-sparing medical management. Nanotherapeutic delivery offers options to stably deliver silencing RNA and microRNA inhibitors to the placenta. Alternatively, targeted delivery of drugs to the fetus may minimize side effects for the mother.

Optimal length and usefulness of temporal artery biopsies in the diagnosis of giant cell arteritis: a 10-year retrospective review of medical records

Biopsy length for diagnosis of giant cell arteritis is debated. We reviewed temporal artery biopsies from a 10-year period in Alberta to identify an ideal length. We suggest a 1.5 cm prefixation length is the optimal length for diagnosing giant cell arteritis.

Serious postoperative complications and reoperation after carpal tunnel decompression surgery in England: a nationwide cohort analysis

Of the 855 832 initial surgeries, 620 procedures led to a serious complication within 30 days after surgery. The study is the largest national study on carpal Tunnel Decompression to date. It provides strong evidence of serious postoperative complications.

Safety and efficacy of abatacept in early diffuse cutaneous systemic sclerosis (ASSET): open-label extension of a phase 2, double-blind randomised trial

Abatacept was well tolerated by patients with early diffuse cutaneous systemic sclerosis. The primary endpoint was modified Rodnan Skin Score at 12 months. Clinically meaningful improvements in mRSS and other outcome measures were observed in both the abatacept and placebo groups.

Clinical criteria for COVID-19-associated hyperinflammatory syndrome: a cohort study

A subset of patients with COVID-19 develops a hyperinflammatory syndrome that has similarities with other hyperinflammatory disorders. We developed a six-criterion additive scale for the development of cHIs. The cHIS scale might be helpful in defining target populations for trials and immunomodulatory therapies.

Anti-C5a antibody IFX-1 (vilobelimab) treatment versus best supportive care for patients with severe COVID-19 (PANAMO): an exploratory, open-label, phase 2 randomised controlled trial

Severe COVID-19 is characterized by inflammation and coagulation in the presence of complement system activation. We aimed to explore the potential benefit and safety of selectively blocking the anaphylatoxin and complement protein C5a with the monoclonal antibody IFX-1.

Otilimab inhibits granulocyte-macrophage colony-stimulating factor (GM-CSF) This is a key driver in immune-mediated inflammatory conditions. We aimed to evaluate the efficacy, safety, and key patient-reported outcomes related to pain in patients with active rheumatoid arthritis.

Risk of hydroxychloroquine alone and in combination with azithromycin in the treatment of rheumatoid arthritis: a multinational, retrospective study

Hydroxychloroquine has received negative publicity for adverse events associated with its authorization for emergency use to treat patients with COVID-19 pneumonia. Long-term use of hydroxychloroquine appears to be associated with increased cardiovascular mortality. The addition of azithromycin appears to increase the risk of heart failure and chest pain or angina.

Long-term hydroxychloroquine use in patients with rheumatic conditions and development of SARS-CoV-2 infection: a retrospective cohort study

Hydroxychloroquine is one of several agents being evaluated in the treatment of severe acute respiratory syndrome coronavirus 2. Overall mortality was lower in the hydroxychloroquine group than in the group that did not receive hydroxychloroquine.

Sirolimus for treatment of patients with inclusion body myositis: a randomised, double-blind, placebo-controlled, proof-of-concept, phase 2b trial

Inclusion body myositis is most frequent in patients older than 50 years. In this pilot study, we aimed to test the efficacy of sirolimus in patients with the disease. The study was a randomized, double-blind, placebo-controlled, proof-of-concept, phase 2b trial. Canker sores were the most frequent side-effect and were mainly mild or moderate in ten patients.
